Digital pathology enables laboratories to achieve increased levels of efficiency, collaboration and throughput. Currently regarded as one of the most promising avenues for the future of research and medicine, whole slide imaging offers one of the most dynamic applications to the pathology workflow. A high-resolution image is created from conventionally prepared histology glass slides and then quickly reviewed and analyzed right at the pathologist’s desktop display. Did you know that failure to prepare could result in trouble during scanning? What are the impacts to your workflow and productivity and the image your pathologists’ views if you are not prepared? In this workshop, we will discuss things to consider when implementing digital pathology, how to choose a digital pathology scanner, and the impact that your processes and procedures have on the quality of images.

Barrett esophagus (BE) represents a premalignant condition characterized by intestinal metaplasia of the distal esophagus, predisposing individuals to the development of esophageal adenocarcinoma. This presentation provides an overview of current practice guidelines for the screening and surveillance of BE, including identification of at-risk populations, optimal surveillance intervals, and endoscopic management strategies based on dysplasia grade. The spectrum of histologic changes in nondysplastic and dysplastic BE are reviewed, as well as challenges faced with interobserver variability between pathologists and the relevance that diagnostic agreement and confirmation has on surveillance and management. Additionally, this presentation explores the evolving role of risk prediction models, novel dysplasia detection tools, and progression-associated biomarkers. This review provides a practical approach to summarizing current screening and surveillance guidelines, differentiating the spectrum of changes in BE and dysplasia, and analyzing the clinical utility of novel risk stratification tools.

"Doing more with less: molecular test optimization and the histology laboratory at the University of Colorado Anschutz Medical Campus"

The Colorado Molecular Correlates (CMOCO) laboratory on the Anschutz Medical Campus offers, among myriad services, molecular profiling for the detection of somatic alterations in solid tumors. Successful testing often hinges on the ability to obtain adequate, high quality nucleic acid from very scant specimens. In this talk, Dr. Young will discuss molecular testing methods, approaches and barriers to testing, and the essential (and ever evolving!) role of the histology laboratory in facilitating successful downstream testing. She will use the experience of the collaboration between the histology lab in the Department of Pathology at the University of Colorado and CMOCO to underscore the significance of the histotechnologist in providing quality patient care in an academic hospital setting.

The spectrum of melanocytic lesions in dermatopathology most frequently includes benign nevi, dysplastic nevi (which are further graded on a scale of mild, moderate, and severe), and melanoma; and accurately differentiating between these is often difficult. As such, immunohistochemical stains are critical in the evaluation and classification of melanocytic lesions. In this presentation, Dr. Preston will cover the basics of the different melanocytic stains such as SOX-10, S100, MART-1/MelanA, HMB45, and PRAME as well as the processing, selection, and clinical utility of each. Finally, this presentation will review how these stains are used by pathologists to aid in the diagnosis of melanocytic lesions (especially determining benign from malignant) and potential pitfalls to their use.
